Yeah, you need to make your file a transparent PNG with no background. I'm not familiar with Painter X, but you need to find a way to save it to PNG and make sure that there is NO white background in the file you're saving to begin with. When programming in the images to the Renpy program what sizes should I use for character art and BG art? Is there a set standard I can go by as far as sizes go??
