Results depend on the person and the effort put into your drawing skills (i.e. how much you practice). Some may take months, some years, some a few weeks.

I suggest you learn anatomy first. Start with the head. Once you perfect drawing the head, then work on torso and down. For ren'ai games, all you really need is the top half of the body. Btw, when I say head, that includes eyes, hair, etc.

I suggest you work on these areas below first. They're what you need to know the most. When you're done with those, work on clothes and the rest of the anatomy.

Head:
Front View
Side View
Quarter View

Hair:
Basics
Spiked
Curly
Straight
Bangs

Other:
Eye Basics
Emotional Expressions

You don't have to use guidelines but it's there for you to keep your face straight or should I say, keep everything positioned correctly. A face is symmetrical and the guidelines reflect that fact. Guidelines help prevent the usual beginner mistakes:
1) The chopped off head. The skull isn't rounded fully and the back or top of the head looks flatter than it should be.
2) Skewed features. Placement of body parts like eyes, nose, and ears are not exactly aligned properly.
3) Low hair cut. The ends of the hair start too close to the eyes resulting in a squashed face.

If you already can eyeball exactly where something goes, then don't use the guidelines but if your work looks horrible without guidelines, then don't be lazy - use the guidelines.

Have you tried drawing your own character head shots yet (with or without the guidelines)?
