I know how to design GUI in ren'py, but is there a way to make GUI using GIMP?

There is, considering the fact that all of the UI's I make are made on GIMP. However, there's no one specific way to make them. You have to come up with your own. A tip I would give you is to look at other game UI's and try to emulate certain elements. That's what I do.

Two ways:

Make an image in GIMP and put hotspots for areas in Ren'py.

Or make template images for buttons and menus that can be scaled and combined to make the GUI on fly in Ren'py. I think this is better since it's more dynamic but for detail backgrounds you may want to combine it with the first method.
