Expressions
For expressions, I find even tiny changes make a big difference. I like to put on the brows, eyes, mouths and fx (blush, gloom or sparkles, etc) on a different layers, so you can mix and match.
(only the eyes change, but it defines the emotion)
(Notice the mouth is
in all the images, but the brows/eyes changes everything)
(Different eyes)

For brows: neutral, furrowed, serious, relaxed/mellow, irritated/confident, angry, sad, raised (happy/surprised), dubious/suggestive
For eyes, the basics are: neutral, narrowed, half, avert, sorrow, sincere, surprised
For mouths, I usually include: 
:] :{ 
:/ :T D: 
- sometimes more, if their personality calls for it, like :3 :V :c etc
FX: <3, blush, gloom, sweat, sometimes other little fx like tears or sparkles, if it suits the character.
If your storyline or character calls for an extreme emotion, I'll be sure to customize and make that work.
