It depends. Sometimes I like to play with girls but some other times (in some games) I need to play as a guy for the storyline and stuff...
I think it depends on the game (and on my mood :D)

I usually choose female, but if I'm playing a game where most of the people are girls, I choose boy to stand out 8) and also if there is like a really hot guy :?.

I prefer a female protagonist but I can play with a male protagonist (Steve-The Flower Shop)as well.

Well... I prefer to play as both in games. I don't see what the big deal is about what gender your character is. I play both equally I suppose... I think I got like that because there are awesome games that you can only play as a male.

I tend to write as the opposite gender (male), so I have no trouble playing as a male.
On the other hand, I also have little trouble playing as a girl. This isn't really a suprise though, considering I identify myself as androgynous (emotionally speaking).

I generally play as whichever gender I feel like playing as.
As an exception though, when I make an MMO account (since they usually let you create more than one character on one account) I make both a male character and a female character.

I'll play both bishoujo games and otome games so I don't care too much either way.

I play as same gender, a lot of people don't, so I wouldn't be bothered by anyone that played as opposite gender.

I always play as female. It just feels weird if I play as male. My brother usually plays as female, which I don't understand why he does that.
I have tried several times playing as male though (like in Pokemon or Harvest Moon) but still, I somehow get a bit distracted by the fact I'm playing not as female.

But I don't really care either way... For Mass Effect, as in your example, I played as both XD

If there's customization and you're not limited to girls via choosing it, I usually play as the cutest guy I can pick. :P
If you are locked into the opposite gender though (depending on how hard I ship them) I'll usually play a girl.

Annnnd for stat purposes, I am a straight girl who doesn't mind BxB. O3O

I don't at all mind playing as the opposite gender.
If there's a choice, I usually play as the girl, though, just becausethey're cuter.
I'm a girl btw for clarification's sake.
If there's major story variation based on the gender/character you play as, I usually play as all the characters unless one (or more) of the other routes is a major turn-off personality-wise or story-wise.

Ideally, I play as female characters, since I'm a girl and I feel I can relate to them more...

... However, as a gay girl, sometimes I do choose to play as male characters if there's a cool female love interest who only a male character can romance. I'm sort of spread all over the place-- GxB games/paths can interest me if I relate to the leading lady, BxG games/paths can interest me if the romanceable girls interest me, BxB only interests me if the plot/setting sounds like something I'd absolutely enjoy romance aside, and GxG is what I yearn for deep in my soul, and very rarely receive (especially since I'm profoundly bored by slice-of-life and that's the majority of GxG fiction)

Games without romance but with gender options, I always choose female. I choose female and then, if I have the opportunity and if it's that kind of game, I turn my lady into a thuggish melee powerhouse who wields improbable weapons. Yeeeah. BD

Same gender (female), every time.
To be honest, it's a pet peeve of mine if a game says CUSTOMISE YOUR CHARACTER!! and doesn't specify if that means I can be male or female, or if I get to choose what kind of beard I give my grizzled white guy.

I remember being the most excited kid in the world when I found out I could play a girl in Pokemon Crystal.

That said, it kinda made me realise how it's common for a game to have a customisable exclusively male character, but I can't think of a game where you can customise a female only character (Feel free to correct me if I'm wrong, in fact I'd like to be proven wrong on this haha).

In Mass Effect and Pokemon, I always chose male, but I have played games with female MC, like Harvest Moon Friends Mineral of Town.
I also played some GxB otome games with female MC.
