Which is better?
I am new new here, and... i dunno if I should mouse draw it right away... i'm still not used to mouse draw, which is better? mouse draw right away or draw it in a paper? if mouse draw what would you recommend??

Based on experience, sketch first in paper, scan, then trace it. If you use Photoshop, the pen tool's your best bet. If you use SAI, you lineart from a linework layer using the pen tool.
I swear, drawing directly in the computer using a mouse will take you a LOT longer, and the result's a bit unsightly (unless you're used on it).

It's really hard to draw with a mouse unless you use vectors/pen lines. Drawing on a paper, scanning and then colouring it gives a much better result.

As for SAI vs PS, it's really up to what you're comfortable with. SAI has pen stabilizers which make freehand lining smoother. PS has a pretty easy to use pen+stroke tool (not sure about SAI but I think there's the same thing). So try it out and see which one suits your needs best.

Paper is definitely easier for me >_>

I find sai is easier to use than photoshop - but i like to combine both ^^'

From a technical angle? Photoshop is a much stronger program. However, it has a lot of features you probably won't use, especially as a beginner, so it all depends on what you feel most comfortable. Do you know any tutorials about coloring pictures/drawings using SAI/photoshop? I want to learn about it.

There are tons of tutorials on DeviantArt on how to colour using Photoshop or even Sai, you should look there. There's a also a lot of videos to be found on YouTube, but then again, most of them are just showing off their drawing process.

You create additional face images showing the emotions, and then copy and paste them onto the 'base' sprite.

The one insight - and I'm not sure if Ren'Py's documentation makes this totally clear - is that there isn't a fundamental difference between an event CG and a background.

You probably won't be very successful drawing on the computer if you can't draw on paper first. A computer program is only a tool to help you draw faster with less mess.
If you have a slower computer, Sai is the way to go. It's a lot cheaper and won't lag on you. There's a free version of Sai called "Portable Sai" which is basically an unlimited version of the Sai trial (which doesn't have all the features that regular Sai has, but, it works just as well)
For the rest of your graphical needs, just get Gimp. It has all the basic filters photoshop has along with some of its own, and, it's free too! I own both photoshop elements and CS5, but I hardly use them besides for filtering/logo making that you can get in Gimp for free.
Also, before asking questions about ren'py, try out the demo game that comes with it and take a look through the documentation here: http://www.renpy.org/doc/html/ There's plenty of info to get you started.
