I keep getting Google Play support is enabled, but build.google_play_key is not defined...
Do I need to be register as a developer to be able to compile my game for Google Play?
If so, once I pay the $25, are there any settings that I need to be aware of?
I tried to add the code below in my options.rpy file but I get an error about expected statement.
build.google_play_key = "..."
build.google_play_salt = ( ... )
I really hope someone can help me because this "Google Play support is enabled, but build.google_play_key is not defined" is driving me insane

You don't need a build key to upload to Google Play. That key is for redeeming purchases and other, monetary uses. Google will give you this if you register for transactions.
You will however, still need to pay the $25USD fee and register a Google Play Developer account regardless of whether your games are free or not.

So, what you are saying is that once I registered on Google Play and paid the $25, I will be able to compile my game without getting the "Google Play support is enabled, but build.google_play_key is not defined..." Am I right?

Pretty much.
What you do for now is leave the # in front of the lines.
When you have paid the Google fees and set up your banking details (so they can pay you your share of sales) they will give you a string that you would put in those lines.
I have not done this personally, so I cannot help much further, sorry.
